The Flash will face off against another villain from the comics in episode 6. UPDATED with casting news.
The Flash have put out a call for their sixth villain. He's not exactly what you would call a legendary rogue, though. In episodes 6 and 7, Barry will face off with Tony Woodward, known to DC Comics fans (or maybe not all that known) as Girder. Via Zap2It we now know that Greg Finley (Star-Crossed) will play Girder.
The character description from CW's casting call (which comes to us via TV Line) says that Woodward is someone from Barry's past, namely, one of the boys who used to give him a hard time as a kid. Now that he has metahuman powers he's "a grinning lunkhead who loves destruction...[and an] unstoppable force…who can transmute any part of his body into solid steel.”
Girder is a creation of Flashexecutive producer Geoff Johns (along with artist Ethan Van Sciver) and first appeared in 2001's The Flash: Iron Heights. He's popped up quite a bit in DC Comics continuity since then.

We're very much into "villain of the week" territory with The Flash, and that's understandable, especially in a show's first season. And Girder may seem like a particularly disposable bad guy. Never mind "seems like," he is. But with 23 episodes likely on the slate, they've gotta save the big/important bad guys for big/important episodes. Look on the bright side, we're going to see a host of supervillains we never expected to see in live-action in the first place this season.
